TheY13-238monoclonalantibodyreactswithhuman,mouse,andratv-H-ras(withinaminoacids120-138)anddoesnotreactwithv-K-ras.v-H-rasbindsGTP/GDPandhasintrinsicGTPaseactivity.RasproteinsalternatebetweenaninactiveformboundtoGDPandanactiveformboundtoGTP,activatedbyaguaninenucleotide-exchangefactor(GEF)andinactivatedbyaGTPase-activatingprotein(GAP).Undernormalconditions,Rasfamilymembersinfluencecellgrowthanddifferentiationevents.MutationsintheRasfamilyofproto-oncogenesareverycommon,beingfoundin20%to30%ofallhumantumors.Raspointmutationsarethesinglemostcommonabnormalityofhumanproto-oncogenes.TheY13-238antibodydoesnotneutralizerasGTPasebindingandhydrolysisinvivoorinvitro.
