TheFGK4.5monoclonalantibodyreactswithmouseCD40alsoknownasBp50.CD40isa48kDatypeItransmembraneglycoproteinthatbelongstothetumornecrosisfactorreceptor(TNFR)superfamily.CD40isexpressedbroadlyonantigen-presentingcells(APCs)suchasdendriticcells,Bcells,macrophages,andmonocytesaswellasnon-immuneendothelialcells,basalepithelialcells,andarangeoftumors.UponbindingtoitsligandCD154,CD40actsasacostimulatorymoleculefortheactivationofBcells,dendriticcells,monocytes,andotherAPCs.CD40playsrolesinBcellactivation,differentiation,proliferationandIgisotypeswitchingaswellasdendriticcellmaturation.AgoNISTicCD40monoclonalantibodieshavebeenshowntoactivateAPCsandpromoteanti-tumorTcellresponses.TheFGK4.5antibodyisanagonisticantibodythathasbeenshowntoactivateCD40expressingAPCs.FGK4.5canalsobeusedtoinhibitCD40/CD154interactioninvitroandinvivo.
