TheYTS177monoclonalantibodyreactswiththemouseCD4.TheCD4antigenisa55kDacellsurfacetypeImembraneglycoproteinbelongingtotheimmunoglobulinsuperfamily.CD4actsasaco-receptorwhichincooperationwiththeTcellreceptor(TCR)interactswithclassIIMHCmoleculesdisplayedbyantigenpresentingcells(APC).CD4isexpressedbythemajorityofthymocytes,mosthelperTcells,asubsetofNK-Tcellsandweaklybydendriticcellsandmacrophages.CD4playsanimportantroleinthedevelopmentofTcellsandisrequiredformatureTcellstofunctionoptimally.TheYTS177antibodyhasbeenshowntocompetewithclonesGK1.5andYTS191forCD4binding.Additionally,theYTS177antibodyhasbeenreportedtobenon-depletingbutbindingdoesinducerapidinternalizationofCD4onbothCD4+Foxp3-TcellsandFoxp3+regulatoryTcells.Further,theYTS177clonehasbeenshowntosuppressorevenpreventallograftrejection,allergicreactionsandautoimmuneresponses.
