Theanti-mouseCD3F(ab")2fragmentistheantigenbindingF(ab")2fragmentsofthe145-2C11monoclonalantibodyonly.ThemajorityoftheFcfragmenthasbeenremovedviapepsindigestion.The145-2C11antibodyF(ab")2fragmentreactswithmouseCD3ε,a20kDatransmembranecell-surfaceproteinthatbelongstotheimmunoglobulinsuperfamily.CD3εisoneoffivepolypeptidechainsthatcombinetoformtheTCRcomplex.CD3εisexpressedonTlymphocytes,NK-Tcells,andtovaryingdegreesondevelopingthymocytes.CD3playsrolesinTCRsignaling,Tlymphocyteactivation,andantigenrecognition.Thecomplete145-2C11antibodyiscommonlyusedtostimulateTcellsinvitrohowever,whenusedinvivoactivationofrestingTcellsresultsincytokinereleaseandtoxicitycausedbyAb-mediatedcross-linkingofTcellsandFcγreceptor-bearingcells.Toavoidthesecomplicationsnon-FcreceptorbindingF(ab’)2fragmentsofthe145-2C11antibodyarecommonlyused.Thisnon-FcR-bindinganti-CD3inducesapoptosisofAg-activatedTcellsinvivo byallowingdurableexpressionoftheTCRandsustainedsignaling.Foxp3+TregshavebeenshowntoberesistanttoCD3antibody-mediateddepletion.
