TheOX-86monoclonalantibodyreactswithmouseOX-40alsoknownasCD134.OX-40isa50kDatypeImembraneglycoproteinandamemberoftheTNFreceptorsuperfamily.OX-40isexpressedonactivatedCD4andCD8Tcells,butisnotfoundonrestingnaïveTcellsormostrestingmemoryTcells.AlthoughitwasoriginallythoughtthatOX-40expressionwasrestrictedtoactivatedconventionalTcells,ithasnowbeenvisualizedonactivatedregulatoryTcells,NKTcells,NKcells,andneutrophils.OX-40playsamajorroleinregulatingbothCD4andCD8Tcellclonalexpansion.Itprovidesacostimulatorysignaltoanantigen-reactingnaiveTcellstoprolongproliferation,aswellasaugmenttheproductionofseveralcytokines.ThisisdemonstratedbyOX-40knockoutmicewhichgeneratefewerprimaryeffectorCD4Tcellsafterimmunization.FurThermore,invivotreatmentwithanagoNISTantibodytoOX-40hasbeenshowntostronglyenhancethegenerationofantigen-specificeffectorTcellsandpreventtheinductionofTcelltolerance.TheOX-86antibodyisanagonisticantibodythathasbeenshowntodelaytumorgrowthinvivo.
