
The145-2C11monoclonalantibodyreactswithmouseCD3ε,a20kDatransmembranecell-surfaceproteinthatbelongstotheimmunoglobulinsuperfamily.CD3εisoneoffivepolypeptidechainsthatcombinetoformtheTCRcomplex.CD3εisexpressedonTlymphocytes,NK-Tcells,andtovaryingdegreesondevelopingthymocytes.CD3playsrolesinTCRsignaling,Tlymphocyteactivation,andantigenrecognition.The145-2C11antibodyhasbeenshowntoinduceTlymphocyteactivation,proliferation,andapoptosisinvitroviabindingandstimulatingtheTCR.WhenusedinvivotheantibodyisreportedtoproduceTcellactivation,anergy,ordeath.ActivationofrestingTcellsinvivoresultsincytokinereleaseandsubsequenttoxicitycausedbyAb-mediatedcross-linkingofTcellsandFcγreceptor(FcR)-bearingcells.Forthisreason,thenon-FcR-binding145-2C11f(ab")2fragmentsaremorecommonlyusedforinvivoapplications.
