TheR6-5-D6monoclonalantibodyreactswithhumanCD54alsoknownasIntercellularAdhesionMolecule-1(ICAM-1),isasingle-chaintransmembraneglycoproteinwithapolypeptidecoreof55kDaandamemberoftheIgsuperfamily.CD54istypicallyexpressedonnon-hematopoieticcellssuchasendothelialcells,thymicepithelialcells,andfibroblastsbutcanalsobeexpressedonmacrophages,T-lymphoblasts,germinalcenterBcellsanddendriticcells.CD54isaligandforLFA-1,areceptorfoundonleukocytes.Whenactivated,leukocytesbindtoendothelialcellsviaCD54/LFA-1andthentransmigrateintotissues.Pro-inflammatorycytokinessuchasIL-1,TNFandIFNγinduceCD54expressiononendothelialcellsandfibroblasts.TheR6-5-D6antibodyhasbeenreportedtoinhibittheinteractionofCD54withleukocytes,therebydecreasingleukocyteadhesiontothevascularendotheliumandinflammatorytissueinjury.
