The1G9monoclonalantibodyreactswithmouseTIGIT(TcellimmunoreceptorwithIgandITIMdomains).TIGITisa26kDa,typeItransmembraneproteinandamemberofthepoliovirusreceptor(PVR)family.TIGIThasbeenfoundtobeexpressedonfollicularThelpercellsinmicewhileinhumansit"sexpressedbymanyTcellsubsetsincludingactivatedTcells,follicularThelpercells,memoryTcells,andregulatoryTcellsaswellasonNKcells.TIGITcaninteractwithcertainmembersofthePVRandPVR-likefamilies,includingPVR,PVRL2,PVRL3,CD155,andCD112.TIGITisthoughttonegativelyregulateNKandTcellactivation.BindingofTIGITonTcellsbydendriticcellsresultsintheirdifferentiationintoatolerogenicphenotype,withincreasedsecretionofIL-10anddiminishedproductionofIL-12.TIGITknock-outmicearemoresusceptIBLetoautoimmunedisease.
