TheB8.2C12monoclonalantibodyreactswithmouseTIM-3(Tcellimmunoglobulinandmucindomain-3)alsoknownasCD366.ThisantibodybindstotheBALB/calleleofTIM-3whilereactivitytotheC57Bl/6alleleissignificantlyweaker.TIM-3isa60kDamemberoftheTIMfamilyofimmunecheckpointreceptorsandexistsasatypeItransmembraneglycoproteinwithamucin-likedomaininitsextracellularportionandatyrosinephosphorylationmotifinitscytoplasmicportion.TIM-3isspecificallyexpressedathighlevelsonthesurfaceofTh1lymphocyteswhereasTh2lymphocytesexpressTIM-1andTIM-2.TIM-3activationoccursviabindingtothecell-associatedC-typelectingalectin-9.UponbindingTIM-3inducesapoptosisofTh1cells.InhibitionofTIM-3signalinginmicehasbeenshowntoexacerbateexperimentalautoimmuneencephalomyelitis,promoteIFNγproductionandTh1cellproliferation.Tim-3hasalsobeenshowntoberequiredfortheinductionoftolerance,asbothTIM-3knockoutanimalsandmicetreatedwithTIM-3-Igfusionproteindisplaydefectsintheinductionofantigen-specifictolerance.Additionally,TIM-3signalingiscurrentlybeingexploredasacancerimmunotherapytargetasCD8TcellswhichexpressbothTIM-3andPD-1exhibitgreaterdefectsinbothcell-cycleprogressionandeffectorcytokineproductionthancellsthatexpressPD-1alone.
