TheMR-1monoclonalantibodyreactswithmouseCD154alsoknownasCD40ligand.CD154existsasa39kDaaccessorymoleculeandbelongstotheTNFsuperfamilyofcytokines.CD154isprimarilyexpressedonthesurfaceofactivatedCD4+Tlymphocytesbutcanalsobeexpressedbyplatelets,mastcells,macrophages,basophils,NKcells,Blymphocytes,CD8+Tlymphocytesaswellasnon-hematopoieticcellsincludingsmoothmusclecells,endothelialcells,andepithelialcells.CD154signalsthroughCD40andisthoughttoplayakeyroleinTandBlymphocytecostimulation.TheMR-1monoclonalantibodyhasbeenreportedtoinhibitinvitroactivationofBlymphocytesbyblockingthebindingofCD154withCD40onThelpercellsaswellasinhibittheformationofgerminalcentersanddisruptantigen-specificTcellresponses.Additionally,theMR-1antibodyblocksinteractionsofTcellsandantigen-presentingcellsinvitroandblocksthedevelopmentofexperimentalautoimmunediseaseinvivo.
