TheXT3.11monoclonalantibodyreactswithmouseTNFα(tumornecrosisfactor-alpha)amultifunctionalproinflammatorycytokine.TNFαexistsasasoluble17kDamonomer,whichformshomotrimersincirculationorasa26kDamembrane-boundform.TNFαbelongstotheTNFsuperfamilyofcytokinesandsignalsthroughitstworeceptors,TNFR1andTNFR2whichcanbeactivatedbyboththesolubletrimericandmembrane-boundandformsofTNFα.TNFαisprimarilyproducedbymacrophagesinresponsetoforeignantigenssuchasbacteria(lipopolysaccharides),viruses,andparasitesaswellasmitogensandothercytokinesbutcanalsobeexpressedbymonocytes,neutrophils,NKcells,CD4Tcellsandsomespecializeddendriticcells.TNFαisknowntoplaykeyrolesinawidespectrumofBIOLOGicalprocessesincludingimmunoregulation,cellproliferation,differentiation,apoptosis,antitumoractivity,inflammation,anorexia,cachexia,septicshock,hematopoiesis,andviralreplication.TNFαdysregulationhasbeenimplicatedinavarietyofdiseases,includingautoimmunediseases,insulinresistance,andcancer.MouseandhumanTNFαshare79%aminoacidsequenceidentityhowever,mouseTNFαisglycosylatedwhilehumanTNFαisnot.TNFαknockoutanimalsdisplaydefectsinresponsetobacterialinfection,characterizedbydefectsinformingorganizedfolliculardendriticcellnetworksandgerminalcenterswithalackofprimaryBcellfollicles.
