The9H10monoclonalantibodyreactswithmouseCTLA-4(cytotoxicTlymphocyteantigen-4)alsoknownasCD152.CTLA-4isa33kDacellsurfacereceptorencodedbytheCtla4genethatbelongstotheCD28familyoftheIgsuperfamily.CTLA-4isexpressedonactivatedTandBlymphocytes.CTLA-4isstructurallysimilartotheT-cellco-stimulatoryprotein,CD28,andbothmoleculesbindtotheB7familymembersB7-1(CD80)andB7-2(CD86).Uponligandbinding,CTLA-4negativelyregulatescell-mediatedimmuneresponses.CTLA-4playsrolesininductionand/ormaintenanceofimmunologicaltolerance,thymocytedevelopment,andregulationofprotectiveimmunity.ThecriticalroleofCTLA-4inimmunedown-regulationhasbeendemonstratedinCTLA-4deficientmice,whichsuccumbat3-5weeksofageduetothedevelopmentofalymphoproliferativedisease.CTLA-4isamongagroupofinhibitoryreceptorsbeingexploredascancertreatmenttargetsthroughimmunecheckpointblockade.The9H10antibodyhasbeenshowntopromoteTcellco-stimulationbyblockingCTLA-4bindingtotheB7co-receptors,allowingforCD28binding.
