TheP84monoclonalantibodyreactswithSignal-RegulatoryProteinα(SIRPα),alsoknownasCD172a.SIRPαisatypeItransmembraneglycoproteinexpressedonmonocytes,macrophages,anddendriticcells.NeuronsandothertissuesofthecentralnervoussystemhavealsobeenshowntoexpressSIRPα.Itsligand,CD47isexpressedbyawidevarietyofcells.SIRPαandCD47regulatedendriticcell-mediatedTcellactivation,neutrophilmigration,andphagocytosis.SIRPαdiffuseslaterallyonthemacrophagemembraneandaccumulatesataphagocyticsynapsetobindCD47whichinhibitsphagocytosisbymacrophages.Anti-SIRPαantibodiesthatblocktheinteractionofSIRPαwithCD47havebeenshowntosuppresstumorformationinmice.TheP84antibodyhasbeenshowntohaveneutralizingactivityinvivoandinvitro.
