TheB6H12monoclonalantibodyreactswithhumanCD47otherwiseknownasintegrin-associatedprotein(IAP),andneurophilin.CD47isanapproximately50kDaglycosylatedfivetransmembraneproteinthatisubiquitouslyexpressedbybothhematopoieticcellssuchasTandBlymphocytes,monocytes,plateletsanderythrocytesandnon-hematopoieticcells.CD47isinvolvedinarangeofcellularprocesses,includingapoptosis,proliferation,adhesion,andmigration.FurThermore,itplaysakeyroleinimmuneandangiogenicresponses.CD47isareceptorforthrombospondin-1(TSP-1),asecretedglycoproteinthatplaysaroleinvasculardevelopmentandangiogenesis.CD47Ishasbeenfoundtobeoverexpressedinmanydifferenttumorcells.Becauseofthis,anti-CD47monoclonalantibodieshavebeenproposedandstudiedasatherapeutictreatmentforhumancancers.TheB6H12antibodyhasbeenshowntoneutralizeCD47andinhibitthegrowthofhepatocellularcarcinomacellsinvitro.
