The528monoclonalantibodyreactswithanepitopeontheextracellulardomainofhumanEGFR(epidermalgrowthfactorreceptor)alsoknownasErbB-1.EGFRisa170kDacell-surfacereceptorandbelongstotheErbBfamilyofreceptors.EGFRsignalingisactivateduponbindingoneofitsligandsincludingepidermalgrowthfactor(EGF),transforminggrowthfactorα(TGFα),Amphiregulin,andheparinbinding-EGF(HB-EGF).Uponactivation,EGFRtransitionsfromaninactivemonomericformtoanactivehomodimer.ThisinitiatesseveraldownstreamsignaltransductioncascadesincludingtheMAPK,AktandJNKpathways,leADIngtoDNAsynthesisandcellproliferation.EGFRoverexpressionorconstitutiveactivationareassociatedwithmanycancers.Forthisreason,anti-EGFRmonoclonalantibodymediatedimmunotherapiesarecurrentlybeingexploredascancertreatments.The528antibodyhasbeenreportedtoblockEGFbindingtoitsreceptorandinhibitA431tumorformationinnudemice.
