The10F.9G2monoclonalantibodyreactswithmousePD-L1(programmeddeathligand1)alsoknownasB7-H1orCD274.PD-L1isa40kDatypeItransmembraneproteinthatbelongstotheB7familyoftheIgsuperfamily.PD-L1isexpressedonTlymphocytes,Blymphocytes,NKcells,dendriticcells,aswellasIFNγstimulatedmonocytes,epithelialcellsandendothelialcells.PD-L1bindstoitsreceptor,PD-1,foundonCD4andCD8thymocytesaswellasactivatedTandBlymphocytesandmyeloidcells.EngagementofPD-L1withPD-1leadstoinhibitionofTCR-mediatedTcellproliferationandcytokineproduction.PD-L1isthoughttoplayanimportantroleintumorimmuneevasion.InducedPD-L1expressioniscommoninmanytumorsandresultsinincreasedresistanceoftumorcellstoCD8Tcellmediatedlysis.Inmousemodelsofmelanoma,tumorgrowthcanbetransientlyarrestedviatreatmentwithantibodieswhichblocktheinteractionbetweenPD-L1andPD-1.The10F.9G2antibodyhasbeenshowntoblocktheinteractionbetweenPD-L1andPD-1andbetweenPD-L1andB7-1(CD80).
