The11B11monoclonalantibodyreactswithmouseIL-4(interleukin-4)amultifunctional14kDacytokine.IL-4isexpressedprimarilybyactivatedTh2cellsandNKcells,andatlowerlevelsbymastcells,andbasophils.IL-4signalsthroughtheIL-4Rα.UponreceptorbindingIL-4stimulatesactivatedBandTlymphocyteproliferation,andthedifferentiationofBcellsintoplasmacells.ItalsoinducesBcellclassswitchingtoIgE,andup-regulatesMHCclassIIproductionwhiledecreasingtheproductionofTh1cells,macrophages,IFNγ,anddendriticcellIL-12.LikeotherTh2associatedcytokines,IL-4isinvolvedintheairwayinflammationobservedinthelungsofpatientswithallergicasthma.The11B11monoclonalantibodyhasbeenshowntoneutralizethebioactivityofnaturalorrecombinantIL-4.
