TheMM27.7B1monoclonalantibodyreactswiththep28subunitofmouseIL-27,amemberoftheIL-6/IL-12cytokinefamily.IL-27isaheterodimericcytokinecomposedtheEBI3(Epstein-Barrvirusinducedgene3)proteinandthep28subunit,alsoknownasIL-30.Myeloidcellsincludingmacrophages,inflammatorymonocytes,microglia,anddendriticcellsarethedominantcellularsourcesofIL-27.ThesecellssecreteIL-27inresponsetoTLRligandsandinflammatorycytokines.IL-27p28signalsthroughIL-27receptors(IL-27R)whicharehighlyexpressedbyNKcellsandactivatedTcellsandtoalesserextentonBcells,naïveTcells,andothers.IL-27signalinghasbeenshowntohavebothpro-inflammatoryandanti-inflammatoryeffects.SignalingthroughIL-27RpromotesTh1polarizationandIFNγproductionbutalsosuppressesthedifferentiationandproliferationofTh2andTh17cellsandinducestheexpressionofIL-10.TheMM27.7B1clonehasbeenshowntoneutralizethebioactivityofmouseIL-27invivoandinvitro andreportedtoalsobindandinhibithumanIL-27.
