The4C9monoclonalantibodyreactswithratbeta2microglobulin(β2M)a12kDacomponentofMHCclassImolecules.β-2-microglobulin,anon-membrane-anchoredglycoprotein,noncovalentlyassociateswiththepolymorphicheavychainofMHCclassImoleculestoformtheHLAclassIantigencomplex.β-2-microglobulinisexpressedonallleukocytes,platelets,endothelialcellsandepithelialcellsandplaysanessentialroleinmediatingproperfoldingandexpressionofMHCclassImolecules.Theimportanceofβ-2-microglobulinisillustratedbyβ-2-microglobulinknockoutmicewhichshowanormaldistributionofTlymphocytes,buthavenomatureCD4orCD8TcellsandarethereforedefectiveinCD4andCD8Tcell-mediatedcytotoxicity.The4C9antibodyrecognizesratβ-2-microglobulinaloneorcomplexedwithFcRnheavychains.
